چکیده

We aimed to evaluate the effect of activated platelet-rich plasma (PRP) on proliferation and chondrogenic differentiation bone marrow-derived mesenchymal stem cells (BMSCs). Six mature male rabbits were included in this study. PRP was obtained by two-step centrifugation from whole blood, it using CaCl2 solution. BMSCs isolated proliferated marrow characterized flow cytometry. Passage 3 cultured high-glucose Dulbecco’s modified Eagle’s medium (HG-DMEM) with four different compositions for consecutive 7 days, including 10% fetal bovine serum, 5% PRP, 15% PRP. Cell counting assays performed cell BMSCs. ( 5 × 10 cells/well 6-well plates) induced conditions 21 days evaluation, commercial (control), (HG-DMEM+5% PRP), (HG-DMEM+10% (HG-DMEM+15% PRP). The gene expression levels ACAN, COL2A1, SOX9 pellets detected. Morphological pathological assessments blind observer. After purifying, percentages CD105(+)/CD34(−) CD44(+)/CD45(−) 96.5% 92.9%, respectively. enhanced all groups, revealed more significant outcome than others day 5. lower three groups control group, but ACAN higher group groups. Histological examinations showed that PRP-treated had regular appearance, larger size, abundant extracellular matrix or still inferior medium. In conclusion, our results show may enhance rabbit However, have limited comparison culture.

The Effect of Platelet Rich Plasma on Chondrogenic Differentiation of Human Adipose Derived Stem Cells in Transwell Culture

Objective(s): Platelet-rich plasma (PRP) has recently emerged as a promising strategy in regenerative medicine due to its multiple endogenous growth factors. Little is known about the role of PRP as a promoter in chondrogenesis of human adipose derived stem cells (hADSCs). The aim of this study was to determine whether PRP may be considered as a natural and easy achievable source o...

Effects of BIO on proliferation and chondrogenic differentiation of mouse marrow-derived mesenchymal stem cells

In vitro expansion of mesenchymal stem cell (MSCs) into large number is necessary for their application in cell-based treatment of articular cartilage defects. On the other hand, some studies have indicated that BIO (6-Bromoindirubin-3-Oxime) possesses mitogenic effects on cell culture. The objective of the present study was to examine the effect of BIO on in vitro expansion and chondrogenic di...

Study of Chondrogenic Effects of Chondrocytes Cocultured With Murine Bone Marrow-Derived Mesenchymal Stem Cells

Purpose: Co-culture systems of marrow derived mesenchymal stem cells (mMSCs) with mature chondrocytes have theoretically been considered as a putative way of MSCs chondrogenic differentiation. MSCs differentiated in this system could be used for transplantation purpose without of any need to their purification since the cells with which MSCs are co cultured are native cartilage cells. Despite o...
